Abstract近年来对抑郁症的节律性研究逐渐开始引起重视。目前临床上对抑郁症的近日节律异常、晨重夜轻的临床症状已有诸多研究,但在动物模型中抑郁样行为的节律性特征和节律基因对动物抑郁样行为节律的影响尚不清楚。本研究采用强迫游泳测试作为抑郁样行为的评价方法,选取ZT2、ZT6、ZT10、ZT14、ZT18、ZT22 作为24h 时的代表时间点,实验对象为野生型C57BL/6 小鼠,mper1 基因敲除小鼠(mper1-/-)、mper2 基因敲除小鼠(mper2-/-)clock 基因敲除小鼠(CLOCK△19),研究野生型小鼠和节律基因敲除小鼠的强迫游泳不动行为的节律性特征,并比较了品系间差异和每个品系的性别差异。通过建立LPS 诱导的小鼠抑郁模型,对外源性刺激引起的抑郁样行为与小鼠内源性抑郁样行为的节律的差别作了初步探索。结果发现,野生型小鼠的强迫游泳不动行为具有明显的节律性特征,mper1-/-小鼠的不动行为的节律性特征被完全改变,而mper2-/-和CLOCK△19 不动行为的节律性特征则完全消失。我们还比较了同种小鼠的雌雄差异,结果发现野生型和mper1-/-小鼠在节律特征和不动时间均有雌雄差异,而mper2-/-和CLOCK△19 仅表现为不动时间的差别。在相同剂量的LPS 作用下,野生型小鼠LPS 诱导抑郁样行为明显增加,而mper2-/-的则没有明显变化。本研究提示了节律基因对强迫游泳不动行为节律以及LPS 诱导的抑郁样行为的重要作用,为未来研究节律基因对抑郁样行为的调节作用提供了基础数据。
Other AbstractIn recent years, the rhythmicity of depression gradually begin to attract attention. In clinic, the diurnal rhythm (being heavy in the morning and light at night) has been reported. But in animal models, the rhythmic characteristics of the depression- like behavior and the influence of clock genes on the rhythm of depression- like behavior is not clear. The forced swimming test was used as an assay method, and ZT2、ZT6、ZT10、ZT14、ZT18 and ZT22 were chosen to be the representative of time points. To study the rhythmic characteristics of immobility behavior of forced swimming test, and to compare the differences between strains or genders, the subjects for the wild type were C57BL / 6 mice, and clock gene knockout mice (mper1-/- , mper2-/-and CLOCK△19). Through the establishment of the depression model induced by LPS in mice, we made a preliminary exploration as the differences in the depression--like behavior caused by exogenous stimulus and endogenous. The results showed that, the immobility behavior of forced swimming test of wild type mice had obvious rhythmic characteristics, which completely changed in mper1-/- and totally lost in both mper2-/- and CLOCK△19 mice. We also compared the differences between male and female mice, and the results showed that both wild type and mper1-/- mice had differences in rhythmic characteristics and the degree of immobility between male and female, while mper2-/- and CLOCK△19 mice only showed the varying levels of the immobility behavior between male and female. Under the same dose, depression behavior-like behavior induced by LPS increased significantly in wild type mice, but there was no obvious changes in mper2-/- mice. This study suggests the important role of the clock genes on the rhythm of immobility behavior of forced swimming test and the LPS-induced depression- like behavior, and providing the basic data for the future research on the regulation of clock genes on rhythmicity of depression-like behavior.
