地塞米松对糖尿病大鼠视网膜血管内白细胞聚集、血管通透性及细胞问黏附因子-1表达的影响

Abstract目的:探讨糖尿病大鼠玻璃体腔注射地塞米松对视网膜血管内白细胞聚集、血管通透性及细胞间黏附因子(ICAM)-1表达的影响。方法:72只Brown—Norway大鼠分为对照组、糖尿病组、糖尿病+生理盐水组和糖尿病+地塞米松组共4组,每组各18只大鼠。除对照组外其余大鼠行链脲佐菌素腹腔注射制造糖尿病模型。糖尿病+生理盐水和糖尿病+地塞米松组大鼠玻璃体腔分别注射10μl生理盐水和10/11地塞米松(5μg/ml)。用吖啶橙血管造影和伊文思蓝方法分别检测白细胞聚集和血管通透性改变情况。以荧光定量多聚酶链式反应和酶联免疫吸附实验检测视网膜中ICAM-1的表达。结果:注射地塞米松后,大鼠视网膜血管内白细胞聚集减少,血管通透性降低,ICAM-1表达减少。4组大鼠视网膜ICAM-1mRNA和蛋白量分别为0.434±0.07、0.764±0.21、0.74±0.18、0.55±0.13和(37.90±4.56)、(76.74±6.68)、(74.32±7.11)、(39.61±4.47)pg/mg。结论:地塞米松能减少糖尿病大鼠视网膜血管内白细胞聚集,降低血视网膜屏障通透性。其机制可能是通过其抑制ICAM-1的表达起作用。
Other AbstractObjective To investigate the effect of intravitreal injection with dexamethasone on leukocyte accumulation, vascular permeability, and the expression of intercellular adhension molecule (ICAM-1) in rats with diabetes. Methods Seventy-two BN rats were divided into 4 groups: control group, diabetes group, diabetes + physiologic saline group, and diabetes+ dexamethasone group, with 18 rats in each group. Streptozotocin was injected into the rats to set up the diabetic model. Accumulated leukocytes were counted in vivo by acridine orange leukocyte fluorography, and retinal vascular permeability was measured by Evans blue assay. The expression of mRNA and protein level of ICAM-1 were evaluated by real-time quantitative polymerase chain reaction analysis and enzymedinked immunosorbent assay. Results In the diabetes+ dexamethasone group, accumulated leukocytes were reduced, retinal vascular permeability decreased, and the expression of ICAM-1 decreased. The expression of ICAM-1 mRNA and protein levels in control group, diabetes group, diabetes + physiologic saline group, and diabetes+ dexamethasone group were 0. 43±0. 07,0. 76±0.21,0.74±0.18,and 0. 55±0. 13; (37. 90±4. 56), (76.74±6.68), (74.32±7.11), and (39.61±4.47) pg/mg respectively. Conclusions Dexamethasone can reduce accumulated leukocytes and retinal vascular permeability, which may be caused by inhibiting the expression of ICAM-1.
